ARGX-111 

ARGX-111 is an afucosylated anti-MET antibody. ARGX-111 blocks HGF-dependent and -independent signaling, downregulating MET expression on the tumor cell surface. ARGX-111 depletes MET-expressing circulating tumor cells through enhanced antibody-dependent cell-mediated cytotoxicity (ADCC), thereby inhibiting tumor metastasis. ARGX-111 depletes circulating tumor cells and inhibits bone and lung metastasis in an orthotopic mouse model of metastatic breast cancer. ARGX-111 is promising for research in breast cancer and other cancers^[1][2].

ARGX-111 可抑制 A549 细胞中 HGF 诱导的 MET 自身磷酸化 (IC₅₀ = 5.3 nmol/L，IMAX = 74%)，也可抑制 MKN-45 细胞 (IC₅₀ = 0.2 nmol/L，IMAX = 63%) 和 EBC-1 细胞 (IC₅₀ = 12.7 nmol/L，IMAX = 69%) MET 自身磷酸化^[1]。
ARGX-111 可中和 HGF 的促迁移作用，抑制 A549 细胞中 78% 的 HGF 依赖性集落形成^[1]。
ARGX-111 (1-10 nM/L) 可抑制 SV40 T 抗原转化的 LOC 人肾上皮细胞球体中 HGF 诱导的分支小管形成^[1]。
ARGX-111 (0.1-100 小时) 可促进 MKN-45、EBC-1 和 MDA-MB 231 细胞中 MET 表达快速降低^[1]。
ARGX-111 (24-48 小时) 不会促进 MET 受体脱落，但可降低 MKN-45 细胞中总的 MET^[1]。
ARGX-111 (0-1000 nM/L) 诱导 MKN-45、NCI-H441 和 A549 细胞发生剂量依赖性裂解^[1]。
ARGX-111 诱导 MKN-45 和 786-O 细胞发生抗体依赖性细胞吞噬作用 (ADCP)^[1]。
