BACE Antibody (YA1914) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to BACE.
研究背景

BACE1 蛋白在淀粉样前体蛋白 (APP) 的蛋白水解过程中发挥着关键作用。它在 A-β 肽序列的 N 末端（特别是 APP 的残基 671 和 672 之间）启动切割，从而产生 β 切割的可溶性 APP，并随后将其释放到细胞外。同时，BACE1 生成相应的细胞相关 C 末端片段，随后由 γ 分泌酶释放。除了其主要靶标外，BACE1 还对 CHL1 表现出切割活性，正如通过其切割该底物的相似性观察到的。

  • Western blot analysis of extracts from Hela(lane 2(20ug) , NIH/3T3(lane 3(20ug) and HEK293(lane 4(20ug) using BACE1 Antibody (HY-P82169) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80438, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.
