DARC Antibody (YA1654) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to DARC.

DARC 是一种非典型趋化因子受体，通过高亲和力结合趋化因子来调控其水平和定位，但这一过程不依赖于经典的配体驱动信号转导级联反应，而是导致趋化因子的隔离、降解或跨细胞转运。它也被称为拦截受体（内化受体）、趋化因子清除受体或趋化因子诱饵受体。DARC 具有广泛的趋化因子结合谱，能与 CXC 和 CC 亚家族的炎症趋化因子相互作用，但不与稳态趋化因子结合。它作为多种趋化因子的受体，包括 CCL2、CCL5、CCL7、CCL11、CCL13、CCL14、CCL17、CXCL5、CXCL6、IL8/CXCL8、CXCL11、GRO、RANTES、MCP-1 和 TARC。DARC 可能通过两种机制调节趋化因子的生物利用度和白细胞募集：在内皮细胞中表达时，它维持组织源性趋化因子从基底侧向管腔侧的跨细胞转运及其向循环白细胞的递呈；在红细胞中表达时，它作为趋化因子的血液储存库和缓冲池，平衡血浆趋化因子水平的潜在波动。此外，DARC 还可作为疟原虫 Plasmodium vivax 和 Plasmodium knowlesi 的受体。

  Western blot analysis of extracts from TF-1 (lane 2(20μg) and TF-1 (lane 3(40μg) using DARC (HY-P81909) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80438, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.

  Flow cytometric analysis of 1X10^6 Hela cells labeling DARC Antibody (YA1654) (HY-P81909, red). Cells were fixed with 4% paraformaldehyde. Then stained with the primary antibody at 1/50 dilution for an hour at 4℃. Alexa Fluor® 488-conjugated AffiniPure Goat Anti-Rabbit IgG H&L (HY-P8002) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Rabbit IgG Isotype Control (HY-P80879, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).
