RAB7 Antibody (YA110) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to RAB7.

RAB7 是一种小 GTP 酶，属于 Rab 家族，作为细胞内膜运输的关键调控因子，参与从运输囊泡形成到与膜融合的全过程。它通过 GDP 结合（失活）和 GTP 结合（激活）状态的循环发挥作用，在激活状态下招募下游效应蛋白，直接调控囊泡形成、运动、锚定和融合。RAB7A 在调控内体-溶酶体运输中起核心作用，包括早期到晚期内体成熟、微管正/负端导向的内体迁移定位，以及通过蛋白互作级联调控内体-溶酶体运输。它还参与生长因子介导的细胞信号传导、营养转运体介导的摄取、神经元轴突中的神经营养因子运输及脂质代谢，并调控黑色素体、病原体诱导的吞噬体（或空泡）和自噬体等特殊内体膜运输。在病原体（如金黄色葡萄球菌和结核分枝杆菌）吞噬体的成熟与酸化中发挥作用，并促进吞噬体与溶酶体融合。与 RAC1 协同调控破骨细胞皱褶边界的形成，控制 NTRK1/TRKA 的内体运输和神经突生长信号传导。此外，它通过调控溶酶体降解影响 EGF-EGFR 复合体的内吞运输，并参与 ADRB2 刺激的脂自噬（一种不依赖胞质脂肪酶的自噬途径，by similar）。它还介导 SDCBP、CD63 和 syndecan 的外泌体释放（PubMed:22660413），以及 ACE2 的囊泡运输和细胞表面表达（PubMed:33147445），并可能参与 PRPH 神经元中间丝组装（by similar）。

• 
  Immunocytochemistry analysis of NIH3T3 cells labeling RAB7 with RAB7 antibody (HY-P80295) at 1/100 dilution. Cells were fixed in 4% paraformaldehyde for 15 minutes at room temperature, permeabilized with 0.1% Triton X-100 for 10 minutes at room temperature, then blocked with QuickBlock™ Blocking Buffer for Immunol Staining for 10 min at room temperature. Cells were then incubated with RAB7 antibody (HY-P80295) at 1/100 dilution in QuickBlock™ Blocking Buffer for Immunol Staining at 4 ℃. Alexa Fluor® 488-conjugated AffiniPure Goat Anti-Rabbit IgG H&L（HY-P8002, Green) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. The Nuclear counterstain was DAPI (Blue).

  Western blot analysis of extracts from Hela (lane 2(20μg), C2C12 (lane 3(20μg), C6 (lane 4(20μg) using CD44 (HY-P80295) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% BSA in TBST for 2 hour at room temperature. The primary antibody (HY-P80295, 1/1000) and Loading control antibody (Beta Actin, HY-P80438, 1/3000) was used in 5% BSA in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (HY-P8004/HY-P8001, 1/10,000) was used for 1 hour at room temperature.
• 
  Flow cytometric analysis of 1X10^6 K562 cells labeling RAB7 Antibody (HY-P80295, red). Cells were fixed with 4% paraformaldehyde and permeabilised with 90% methanol. Then stained with the primary antibody at 1/50 dilution for an hour at 4℃. Alexa Fluor® 488-conjugated AffiniPure Goat Anti-Rabbit IgG H&L (HY-P8002) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Rabbit IgG Isotype Control (HY-P80879, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).
