LysoTracker Blue DND-22 

LysoTracker blue DND-22 is a blue fluorescent dye (Ex/Em: 373/422 nm). LysoTracker blue DND-22 stains acidic regions in living cells. LysoTracker blue DND-22 is used in the researches of neurodegenerative diseases and leukemia^[1][2].

染色步骤^[2]
1) 在 37°C 和 5% CO₂ 的条件下，将 U937 细胞培养在含有 10% fetal bovine serum (HY-T1000)、100 单位/mL Penicillin 和 50 μg/mL Streptomycin (HY-B1906) 的 IMDM 培养基中。
2) 用标记的 oxLDL (18 μg/mL) 或 oxLDL-IC (24 μg/mL) 处理细胞。
3) 在与标记的 oxLDL 和 oxLDL-IC 孵育结束前 30 min，将 50 nM LysoTracker Blue DND-22 加入细胞中。
4) 孵育结束后，用无血清 IMDM 洗涤细胞三次，以去除未结合的染料。
5) 将细胞悬浮于 100 μL IMDM 中，然后将 10 μL 等分试样装入玻璃毛细管中并密封。
6) 使用共聚焦显微镜观察细胞，激发波长为 373 nm，发射波长为 422 nm，以观察溶酶体定位。
