α-Casein (90-95) is a partial agonist of opioid receptors and a copper ion ligand, with opioid activity. α-Casein (90-95) inhibits the secretion of β-hexosaminidase by rat peritoneal mast cells (PMC) with IC50= 0.1 μM. α-Casein (90-95) inhibits the proliferation of prostate cancer cells LNCaP, DU145, and PC3 with IC50 of 0.94 nM, 137 nM, and 6.92 nM, respectively. α-Casein (90-95) activates Gi-like proteins through a membrane-assisted, receptor-independent pathway, or reversibly binds to opioid receptors, inducing intracellular calcium release and conformational changes, and exerts the activity of promoting mast cell secretion and inhibiting tumor cell proliferation. α-Casein (90-95) can be used in the study of the mechanisms of allergic diseases and prostate cancer[1][2][3].

ELISA Assay[2]

Cell Line: Rat peritoneal mast cells (PMC)
Concentration: 0.1 mM (secretagogue assay), 0.5 mM (BAC inhibition assay)
Incubation Time: 10 min (secretion), 5 min (BAC preincubation)
Result: Induced 15.2% β-hexosaminidase release (vs. 5.5% spontaneous release) at 0.1 mM, with maximal secretion within 1 min.
Preincubation with 3.0 μg/mL Benzalkonium chloride (HY-B2232) abolished secretion (inhibition 50.1%), indicating membrane-mediated Gi-protein activation.

2.  If water fails, try dissolving the peptide in a 10%-30% acetic acid solution.
3.  If the peptide still does not dissolve, try dissolving the peptide in a small amount of DMSO.
Zero (=0) 1.  Try to dissolve the peptide in organic solvent (acetonitrile, methanol, etc.) first.
2.  For very hydrophobic peptides, try dissolving the peptide in a small amount of DMSO, and then dilute the solution with water to the desired concentration.
