MAdCAM1 蛋白, Human (HEK293, His)

MAdCAM1 Protein is an important cell adhesion molecule and a member of the immunoglobulin superfamily. MAdCAM1 Protein plays a key role in immune system cell migration and mucosal immune responses. MAdCAM1 Protein, Human (HEK293, His) is a recombinant MAdCAM1 protein expressed by HEK293, with a C-6*His tag and glycosylation modification^[1].

MAdCAM1 是由内皮细胞表达的跨膜糖蛋白，主要位于肠道相关淋巴组织血管内皮及慢性炎症部位血管。MAdCAM1 通过与淋巴细胞表面整合素 α4β7 结合，介导淋巴细胞归巢至肠道黏膜组织及炎症状态下的肝脏等组织。MAdCAM1 在肠道炎症或感染时表达上调以促进淋巴细胞浸润增强局部免疫防御，其异常表达与炎症性肠病密切相关^[1]。

PF-00547659 可与 MAdCAM1 蛋白, Human 结合，并在体外抑制 α4β7 表达细胞与人和食蟹猴 MAdCAM 的粘附^[2]。

1.Measured by the ability of the immobilized protein to support the adhesion of Jurkat human T-lymphocyte leukemia cells in the presence of 1 mM MnCl2. The ED₅₀ for this effect is 1.987 μg/mL, corresponding to a specific activity is 503.2 units/mg.
2.Measured by the ability of the immobilized protein to support the adhesion of Jurkat human T-lymphocyte leukemia cells. When 5×10⁴ cells/well are added to Human MAdCAM-1 coated plates (2.5 μg/mL with 100 µL/well), approximately 52.575%-62.728% will adhere after 1 hour incubation at 37 °C in the presence of 1 mM MnCl₂.

It is not recommended to reconstitute to a concentration less than 100 μg/mL in ddH₂O. For long term storage it is recommended to add a carrier protein (0.1% BSA, 5% HSA, 10% FBS or 5% Trehalose).

Stored at -20°C for 2 years from date of receipt. After reconstitution, it is stable at 4°C for 1 week or -20°C for longer (with carrier protein). It is recommended to freeze aliquots at -20°C or -80°C for extended storage.
