MIP-3 alpha/CCL20 蛋白, Human (His)

MIP-3 alpha/CCL20 Protein, Human (His) is a CC chemokine that attracts lymphocytes and mild neutrophils by binding to and acting on the chemokine receptor CCR6. It induces intracellular calcium mobilization and mediates cancer, various autoimmune diseases, and antimicrobial effects. MIP-3 alpha/CCL20 Protein, Human (His) is a recombinant human CCL20 (A27-M96) expressed by E. coli  with six-His tags at the N-terminus^[1].

CCL20，也称为巨噬细胞炎症蛋白3α（MIP-3-α）和肝激活调节趋化因子（LARC），是属于CC趋化因子家族的一种小细胞因子，位于人类基因组中的2号染色体上。它在胸腺、睾丸、前列腺和肠道中表达水平较低。CCL20 与 CCR6 结合并充当趋化因子受体，并与 CCR6 协同抑制 CD8 T 细胞的功能。CCL20 具有抗菌活性，与炎症性肠病、类风湿性关节炎、牛皮癣等自身免疫性疾病以及肿瘤疾病有关。它可以诱导Treg细胞侵入肿瘤组织，募集树突状细胞促进免疫抑制，并诱导内皮细胞增殖和新内膜形成^[1][2]。

CCL2 (human, -5 μg/mL) inhibits B. abortus 238 with the LD₅ (the dose that achieves 5% reduction of CFU) > 5 μg/mL, effects on B. abortus RB51 in a dose-dependent manner with a LD₅ of 8.7 μg/mL and demonstrates anti-E. coli activity with a LD₅ of 1.5 μg/mL^[3].
CCL2 is up-regulated in human colon adenocarcinoma cell lines stimulated by pro-inflammatory cytokines IL-1α and TNF-α and can function as an NF-κB target gene^[4].

Measured by its ability to chemoattract THP-1 human acute monocytic leukemia cells. The ED₅₀ for this effect is 8.662 ng/mL, corresponding to a specific activity is 1.154×10⁵ U/mg.

It is not recommended to reconstitute to a concentration less than 100 μg/mL in ddH₂O. For long term storage it is recommended to add a carrier protein (0.1% BSA, 5% HSA, 10% FBS or 5% Trehalose).

Stored at -20°C for 2 years from date of receipt. After reconstitution, it is stable at 4°C for 1 week or -20°C for longer (with carrier protein). It is recommended to freeze aliquots at -20°C or -80°C for extended storage.
