SLC5A1 是一种电生性 Na(+) 偶联糖同向转运蛋白，主动在质膜上转运 D-glucose 或 D-galactose，其 Na(+) 与糖的偶联比例为 2:1。转运活性由 Na(+)/K(+) 泵建立的跨膜 Na(+) 电化学梯度驱动。主要功能是将膳食单糖从肠上皮细胞转运至血液，负责 D-glucose 或 D-galactose 在肠上皮细胞顶侧刷状缘膜上的吸收，而基底外侧的排出则由 GLUT2 提供。此外，在肠内分泌细胞中作为 D-glucose 传感器，触发控制食物摄入和能量稳态的肠促胰岛素 GCG 和 GIP 的分泌（By similarity）。与 SGLT2 一起在肾小球滤液中重吸收 D-glucose，在近端小管 S3 段中发挥非冗余作用（By similarity）。将 D-glucose 转运至子宫内膜上皮细胞，控制糖原合成和为胚胎提供营养支持，以及在受孕前子宫内膜的蜕膜化转变。作为水通道，能够在糖和 Na(+) 摄取时响应渗透梯度，实现被动跨膜水转运，具有与水通道蛋白相当的高水导性，因此预计在上皮水通透性中发挥重要作用，尤其是在小肠中。
