IL-12 beta 蛋白, Rat (HEK293, His)

IL-12 beta Protein, also known as natural killer cell stimulatory factor 2, is a common subunit (p40) of IL-12 and IL-23. IL-12 is a inflammatory factor expressed by activated macrophages, and involves in Th1-type immune response against cancer^[1]. IL-12 beta Protein located outside the cell membrane, involves in singalling mediated by Jak-STAT^[3]. IL-12 beta Protein consists of 335 amino acids (M1-S335) with a lg-like domain (43-90 a.a). IL-12 beta Protein, Rat (M1-S335) is produced in HEK293 cells with a C-Terminal His-tag.

IL-12β蛋白，又称IL12 p40亚基或IL-12B，与IL-12 p35亚基（IL-12A）异二聚化形成IL-12，与IL23 p19亚基异二聚化形成IL-23，发挥不同的调节作用功能^[1]。
IL-12 和 IL23 属于 IL-12 家族，参与促炎反应，由活化的巨噬细胞表达，是 Th1 细胞发育的重要诱导剂^[2]。
IL-12 信号通过 p-STAT4 介导的 IL-12Rβ1 和 IL-12Rβ2 传递，而 IL-23 信号通过 p-STAT1 介导的 IL-12Rβ1 和 IL-23R 传递p-STAT3^[3]。
大鼠IL-12 β α蛋白的氨基酸序列与人类差异很大（69.04%），但与小鼠的相似性较高（92.24） %).
白细胞介素 12 (IL-12) 家族已知是一种炎症因子，可诱导自身免疫性炎症，因此可能是自身免疫性炎症性疾病的原因，并且可能对肿瘤发生很重要^[4]。

IL-12/rAAV2 (encoding IL-12 in rat) (1.96 × 112 particles/mL, 3 μL; i.c.v.; 5 μL/min) efficiently induces long lasting expression of IL-12 in rats, and results the greater infiltration of activated microglia cells in the tumor associated improved immune reactions, leading the inhibited growth of implanted glioblastoma and the increased survival time of rats^[5].

IL-12 level is reduced in white adipose tissue (WAT, involved in chronic diseases such as obesity, type 2 diabetes, and atherosclerosis) compared with skeletal muscle (SM) in intense exercise training and weight loss rat^[6].

Measured by its binding ability in a functional ELISA. Immobilized rat IL12B at 10 μg/mL (100 μl/well) can bind mouse IL12RB2 and the EC₅₀ is 0.6-1.5 μg/mL.

It is not recommended to reconstitute to a concentration less than 100 μg/mL in ddH₂O.

Stored at -20°C for 2 years. After reconstitution, it is stable at 4°C for 1 week or -20°C for longer (with carrier protein). It is recommended to freeze aliquots at -20°C or -80°C for extended storage.
