USP10-IN-4 is a potent ubiquitin-specific protease 10 (USP10) inhibitor with an IC50 of 10.87 μM and a Kd of 365 nM. USP10-IN-4 effectively inhibits USP10-mediated proteasomal degradation of downstream proteins YAP and p53, leading to the subsequent downregulation of CDK4 in the p53 signaling pathway. USP10-IN-4 promotes apoptosis in HCC cells and inhibits the onset and progression of liver cancer. USP10-IN-4 can used for the study of hepatocellular carcinoma (HCC)[1].

Cell Cycle Analysis[1]

Cell Line: Huh-7 cells
Concentration: 5, 20 and 40 μM
Incubation Time: 24 h
Result: Increased the proportion of cells in the G1 phase from 45% to 68%.

Western Blot Analysis[1]

Cell Line: Huh-7 cells
Concentration: 0, 0.5, 1, 5, 10 and 20 μM
Incubation Time: 0, 6, 12, 24, 36, 48 h
Result: Reduced the expression of YAP and p53 in a time- and concentration-dependent manner.
Reduced CDK4, and Cyclin B2 proteins.
