4-Nitrophenyl phosphate (p-Nitrophenyl phosphate) is widely used as a small molecule phosphotyrosine-like substrate in activity assays for protein tyrosine phosphatases.4-Nitrophenyl phosphate is a colorless substrate that upon hydrolysis is converted to a yellow 4-nitrophenolate ion that can be monitored by absorbance at 405 nm[1].

体外研究
(In Vitro)

1. 开封前，请将 4-Nitrophenyl phosphate 平衡至室温。
2. 将 10 mg 4-Nitrophenyl phosphate 溶于 10 mL 1X Diethanolamine (HY-W035903) 底物缓冲液中。
注意：二乙醇胺的有效浓度为 10 mM 至 1 M。
3. 在每个微孔板孔中加入 100 μL 4-Nitrophenyl phosphate 溶液。
4. 将微孔板在室温下孵育 30 分钟，或直至显色。
5. 每孔加入 50 μL 2 N NaOH 终止反应。
6. 测量 405 nm 处的吸光度。或者，使用动态 ELISA 读数仪监测反应。
