Cullin 4A/4B Antibody (YA2583) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to Cullin 4A/4B.

Cullin 4A/4B 是多种 cullin-RING 类 E3 泛素-蛋白连接酶复合物的核心组分，通过介导靶蛋白的泛素化及后续的蛋白酶体降解发挥作用。其功能特异性取决于可变的底物识别亚基，其中 CUL4B 可能在复合物中充当支架蛋白，通过定位底物和泛素结合酶促进催化反应。该复合物参与辐射诱导 DNA 损伤后 CDT1、组蛋白 H2A、H3 和 H4 的多聚泛素化，并通过 DDB2 靶向 UV 损伤染色质，可能对 DNA 修复和复制至关重要。部分 DCX 复合物（含 TRPC4AP 或 DCAF12 作为底物识别组分）属于 DesCEND 通路，可识别位于靶蛋白 C 末端的降解信号（C-degron），导致其泛素化和降解。DCX(AMBRA1) 复合物通过介导磷酸化细胞周期蛋白 D（CCND1/2/3）的泛素化调控 G1 向 S 期转换，并作为 CRL5 复合物的调节因子降解 Elongin-C（ELOC）。此外，Cullin 4A/4B 为细胞周期蛋白 E（CCNE1/2）泛素化所必需，影响 G1 期进程，同时通过依赖 26S 蛋白酶体的机制与 MLST8 相互作用调控 mTORC1 通路。与 CUL4A 共同参与核糖体生物合成。

  Western blot analysis was performed on extracts from Hela (lane 1, 15 μg), 293 (lane 2, 15 μg), 3T3 (lane 3, 15 μg), and Jurkat (lane 4, 15 μg) using Cullin 4A/4B Rabbit mAb.Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST at 4°C overnight.The primary antibody (1:1000 dilution) and the loading control antibody (beta-Tubulin, HY-P80487, 1:60000 dilution) were incubated in 5% non-fat milk in TBST for 1 hour at 37°C.Goat Anti-Rabbit IgG-HRP Secondary Antibody (1:20000 dilution) was then applied for 40 minutes at 37°C.
