NCOA4 Antibody (YA4631) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to NCOA4.
研究背景

NCOA4 是一种货物受体，负责铁结合 ferritin 复合体的自噬周转，在铁稳态中发挥核心作用。它作为适配器将 ferritin 递送至溶酶体并进行自噬降解，这一过程称为 ferritinophagy。在饥饿或铁耗竭条件下，NCOA4 将 ferritin 复合体靶向至自噬溶酶体。它可能通过调节 hemin 诱导的红系分化来确保高效的红细胞生成。部分研究表明，NCOA4 能增强雄激素受体 AR 的转录活性，并作为过氧化物酶体增殖物激活受体 (PPAR) gamma 的配体非依赖性共激活因子；但另一项研究显示其对 AR 的共激活作用较弱，且不改变 AR 的配体响应性。NCOA4 还能结合 DNA 复制起始位点，这种结合不限于活跃转录位点，可能独立于核受体转录共激活功能，并通过与 MCM2-7 复合体相互作用阻碍 DNA 解旋，从而抑制 DNA 复制起始的激活。
