Propidium monoazide 

Propidium monoazide is a photoreactive DNA-binding dye that preferentially binds to dsDNA. Propidium monoazide (PMA) prevents DNA from dead bacteria from being amplified during the PCR. PMA-PCR enhanced both the specificity and the sensitivity of PCR^[1]. Propidium monoazide is a click chemistry reagent, it contains an Azide group and can undergo copper-catalyzed azide-alkyne cycloaddition reaction (CuAAc) with molecules containing Alkyne groups. It can also undergo strain-promoted alkyne-azide cycloaddition (SPAAC) reactions with molecules containing DBCO or BCN groups.

1. Propidium monoazide 标记细菌
1) 在昏暗的房间中将 5 µL 2 mM 光反应染料 Propidium monoazide 添加到 500 mL 样品中，最终浓度为 20 μM/µL。

注：计算循环阈值 Ct 差值 (ΔCt)，其中：ΔCt 活菌是经和未经 PMA 处理的活菌 Ct 值之间的差值，ΔCt 死菌是经和未经 PMA 处理的死菌 Ct 值之间的差值。理想的 PMA 处理条件应具有最高的 ΔCt 死菌和最低的 ΔCt 活菌。
