IL-22 由多种 T 细胞、ILC3、中性粒细胞和巨噬细胞等细胞产生，IL-22 有一种由 IL-22R1 和 IL-10R2 组成的特异性受体，存在于多种器官的非免疫细胞上。IL22RA1 与 IL22 结合诱导酪氨酸激酶 JAK1 和 TYK2 活化，激活 STAT3 和 p38 MAPK，进而通过 STAT3、ERK1/2、MAPK 和 PI3K/AKT 通路促进细胞存活和增殖。IL-22 也促进 GSK3B 在 'Ser-9' 和 CTTN 的磷酸化^[5]。
IL-22 受到 IL-23、IL-1β、IL-7、AhR、Notch 等细胞因子的正调控，受到 IL-22BP、TGF-β、IL-27、ICOS、c-Maf、IL-25 等细胞因子的负调控^[6]。
