SiR-Halo is a fluorescent probe based on silicon-containing rhodamine derivatives, which can be used for multicolor imaging of proteins in live cells (Ex/Em = 652/668 nm)[1].

体外研究
(In Vitro)

1. 溶液配制[1]
1.1 储备液配制
溶剂：DMSO
浓度：1-10 mM (根据实验优化)。
储存：分装后避光保存于 -20°C 或 -80°C。避免反复冻融。
1.2 工作液配制
用 PBS 或无血清培养基稀释至 1-10 µM。
注意：工作液应立即配制并使用，并避光保存。

2. 细胞染色
2.1 用合适的构建体转染表达 EGFP-α-微管蛋白和 H2B-mCherry 的 HeLa 细胞。
2.2 底物在成像前无需清洗 (1.5 µM SiR-Halo)。
2.3 充分混匀，37°C 孵育 30 分钟。
2.4 荧光显微镜检测 (Ex/Em = 652/668 nm)。

  • Do most proteins show cross-species activity?

    Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
