SUMO 蛋白酶能够识别并且高效地将 SUMO (Small Ubiquitin-Like Modifier) 从融合蛋白上切割下来。相对于 EK 和 TEV 等蛋白酶的短小识别位点，SUMO 蛋白酶能够识别完整的 SUMO 结构，所以 SUMO 蛋白酶不会错切融合的蛋白。SUMO 蛋白酶的效力可维持在较宽范围的反应环境体系中，例如温度 (4-30℃)、pH (5.5-9.5) 等。SUMO 蛋白酶还具有多聚His标签，便于融合蛋白切割后的亲和层析纯化。
使用说明
为达到最好的酶切效果，确保重组蛋白为部分或完全纯化的蛋白。
对于大部分融合蛋白，SUMO 蛋白酶最理想的反应液中 NaCl 的浓度为 150 mM。然而根据实际情况可在 100 mM-300 mM 之间调节 NaCl 的浓度以达到最佳的效果，也需要考虑到酶中盐的浓度 (终浓度为 12.5 mM) 和底物中盐的浓度。咪唑的浓度应低于 150 mM，若高于该浓度会影响 SUMO 蛋白酶的活性。
注意：
1) 不同蛋白底物，SUMO 蛋白酶的酶切效率会有差异。建议在进行大量酶切前，先使用小量酶测试其不同温度下的酶切效率。
2) -20℃ 储存，避免反复冻融。
