ALKBH5 Antibody (YA3429) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to ALKBH5.

ALKBH5 是一种特异性去甲基化 N(6)-methyladenosine (m6A) RNA 的双加氧酶（dioxygenase），m6A 是高等真核生物信使 RNA (mRNA) 中最常见的内部修饰（by similar）。该酶通过氧化去甲基化作用（需要分子氧、α-酮戊二酸和铁作为辅因子）调控 RNA 的 m6A 修饰，进而影响 mRNA 的加工、翻译和出核过程（by similar）。ALKBH5 也能在体外对单链 DNA 中的 N(6)-methyladenosine 进行去甲基化（by similar）。在精子发生过程中，ALKBH5 通过介导精母细胞和圆形精子细胞中 m6A 的去甲基化，确保靶转录本的正确剪接和产生更长的 3’-UTR mRNA，从而支持减数分裂后期和单倍体阶段的正常发育（by similar）。此外，ALKBH5 通过液-液相分离参与核内无膜细胞器 paraspeckle 的组装，该过程与 NEAT1 非编码 RNA 等转录本的 m6A 去甲基化偶联（by similar）。在免疫调控方面，ALKBH5 通过去甲基化 JAG1 和 NOTCH2 转录本抑制 γ-δ T 细胞增殖，并通过介导 CCL28 mRNA 的去甲基化和去稳定化抑制调节性 T 细胞 (Treg) 的募集（by similar）。

  Immunohistochemical analysis of paraffin-embedded mouse testis tissue using ALKBH5 Antibody. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0) for 8 minutes. The tissues were blocked in QuickBlock for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody at 1/100 dilution in 4℃ overnight. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.
