HuR 抗体 (YA1933)

HuR Antibody (YA1933) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to HuR.

HuR 是一种 RNA 结合蛋白，能够结合 mRNA 的 3'-UTR 区域并增强其稳定性。它参与胚胎干细胞 (ESC) 分化过程：优先结合未被 N6-甲基腺苷 (m6A) 甲基化的 mRNA，稳定这些 mRNA 并促进 ESC 分化 (By similarity)。此外，它也能结合含 m6A 的 mRNA，例如通过结合含 m6A 的 MYC mRNA 来维持 MYC 的稳定性。HuR 能够结合目标 mRNA 3'-UTR 中的 poly-U 元件和 AU 富集元件 (AREs)，例如 FOS 和 IL3/interleukin-3 mRNA 中的 ARE。在 FOS 的 ARE 中，它结合一个包含 AUUUA、AUUUUA 和 AUUUUUA 基序的 27 核苷酸核心元件，并在体外优先结合 5'-UUUU[AG]UUU-3' 基序。与 ZNF385A 共同作用时，HuR 结合 p53/TP53 mRNA 的 3'-UTR 以调控其受 CDKN2A 诱导的核输出，从而可能调节 p53/TP53 表达并部分介导 CDKN2A 的抗增殖活性 (By similarity)。此外，它还能提高含有 ARE 的 leptin mRNA 的稳定性。

  Immunohistochemical analysis of paraffin-embedded rat brain tissue using HuR Antibody. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0) for 8 minutes. The tissues were blocked in QuickBlock for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody at 1/100 dilution in 4℃ overnight. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.
