RICTOR Antibody (YA4339) is a Mouse-derived and non-conjugated IgG1 monoclonal antibody, targeting to RICTOR.

RICTOR 是 mechanistic target of rapamycin complex 2 (mTORC2) 的关键组分，该复合物通过生长因子信号调控细胞增殖、细胞骨架组织、脂肪生成及合成代谢等通路。mTORC2 可磷酸化并激活 AGC 蛋白激酶家族成员（如 AKT、PKC 和 SGK1），且不同于 mTORC1，其对营养状态不敏感。RICTOR 在复合物中可能作为分子适配器，并赋予 mTORC2 对 FKBP12-rapamycin 的耐药性。mTORC2 通过介导 AKT1 不同位点的磷酸化（如 Thr-450、Ser-473 等）促进其完全激活，同时还能磷酸化 SGK1 (Ser-422) 和 PRKCA (Ser-657)（by similar）。此外，该复合物参与胰岛素信号（如磷酸化 FBXW8 和 IGF2BP1）（by similar）及通过 Rho GTPases 调控肌动蛋白细胞骨架。

• 
  Western blot analysis of extracts from Hela (lane 2(20μg), HepG2 (lane 3(20μg) and HEK293(lane 4(20μg) using RICTOR (HY-P84642) Mouse mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80438, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.

  Immunocytochemistry analysis of NIH/3T3 cells labeling RICTOR with RICTOR antibody (HY-P84642) at 1/500 dilution. Cells were fixed in 4% paraformaldehyde for 15 minutes at room temperature, permeabilized with 0.1% Triton X-100 for 10 minutes at room temperature, then blocked with QuickBlock™ Blocking Buffer for Immunol Staining for 10 min at room temperature. Cells were then incubated with RICTOR antibody (HY-P84642) at 1/500 dilution in QuickBlock™ Blocking Buffer for Immunol Staining at 4 ℃. Alexa Fluor® 488-conjugated AffiniPure Goat Anti-Mouse IgG H&L（HY-P8005, Green) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. The Nuclear counterstain was DAPI (Blue).

Cells were fixed in 4% paraformaldehyde for 15 minutes at room temperature, permeabilized with 0.1% Triton X-100 for 10 minutes at room temperature, then blocked with QuickBlock™ Blocking Buffer for Immunol Staining for 10 min at room temperature. Cells were then incubated with RICTOR antibody (HY-P84642) at 1/200 dilution in QuickBlock™ Blocking Buffer for Immunol Staining at 4 ℃. Alexa Fluor® 488-conjugated AffiniPure Goat Anti-Mouse IgG H&L（HY-P8005, Green) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. The Nuclear counterstain was DAPI (Blue).

  Immunohistochemical analysis of paraffin-embedded Mouse liver tissue using RICTOR antibody. The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0) for 8 minutes. The tissues were blocked in QuickBlock for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (HY-P84642, 1/500) in 4℃ overnight. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.
