SP1 Antibody (YA064) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to SP1.

SP1 是一种转录因子，能够响应生理和病理刺激激活或抑制转录。它以高亲和力结合富含 GC 的基序，并调控大量参与细胞生长、凋亡、分化和免疫反应等过程的基因表达。SP1 受到多种翻译后修饰（如磷酸化、SUMO 化、蛋白水解切割、糖基化和乙酰化）的高度调控。它还能结合 PDGFR-alpha G-box 启动子，可能在调节细胞对 DNA 损伤的反应中发挥作用，并参与染色质重塑。SP1 在调控 FE65 基因表达中起关键作用，与 ATF7IP 形成复合物后，通过诱导 TERT 和 TERC 基因表达维持癌细胞的端粒酶活性。其亚型 3 的转录激活能力比亚型 1 更强，还能正向调控核心时钟组件 BMAL1 的转录。此外，SP1 在招募 SMARCA4/BRG1 至 c-FOS 启动子中发挥作用。通过相似性研究（By similarity）发现，SP1 在脑损伤后通过调控 RNF112 的表达保护细胞免受氧化应激。

  Western blot analysis of extracts from Ramos (lane 1) and THP-1 (lane 2) and 293T(lane 3) and HeLa (lane 4) using SP1 antibody. Proteins were transferred to a PVDF membrane and blocked with 5% nonfat powdered milk in PBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (GAPDH, 1/3000) was diluted with 5% nonfat powdered milk in PBST at 4°C overnight. Goat Anti-Rabbit IgG-HRP Secondary Antibody (1/8,000) was incubated for 45min at room temperature.
