TXNIP Antibody (YA024) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to TXNIP.

TXNIP 是一种氧化应激介质，通过抑制 thioredoxin 活性或限制其生物利用度发挥作用。它与 COPS5 相互作用，恢复 COPS5 诱导的 CDKN1B 稳定性抑制，并阻断 COPS5 介导的 CDKN1B 从细胞核到细胞质的转运（By similarity）。TXNIP 还作为转录抑制因子，可能通过充当转录因子和核心抑制复合物之间的桥梁分子发挥作用，其过表达会诱导 G0/G1 细胞周期停滞。此外，TXNIP 是自然杀伤细胞成熟所必需的（By similarity），并作为肿瘤细胞生长的抑制剂。它还能抑制 DDIT4 的蛋白酶体降解，从而有助于抑制哺乳动物雷帕霉素靶蛋白复合物 1（mTORC1）。

• 
  Immunohistochemical analysis of paraffin-embedded Mouse kidney tissue using TXNIP Antibody. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0) for 8 minutes. The tissues were blocked in QuickBlock for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (HY-P80361, 1/1000) in 4℃ overnight. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

  Western blot analysis of extracts from Hela (lane 2(20μg) or lane 3(40μg)) and NIH3T3 (lane 4(20μg) or lane 5(40μg)) using TXNIP (HY-P80361) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% BSA in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80438, 1/10000) was used in 5% BSA in TBST at 4°C overnight. Goat Anti-Rabbit IgG-HRP Secondary Antibody (1/10,000) was used for 1 hour at room temperature.
