CellTracker Blue CMAC (solution) 

CellTracker Blue CMAC (solution) is a non-fluorescent cell membrane permeable dye. The chloromethyl groups of CellTracker Blue CMAC are enzymatically cleaved by intracellular glutathione (GSH) to generate a fluorescent product (blue fluorescence, Ex/Em: 360/460 nm). CellTracker Blue CMAC is suitable for long-term cell tracking (up to 72 hours) and cell proliferation studies, and can also quantify GSH levels^[1][2][3].
Solvent and concentration: DMSO: 10 mM

操作步骤^[2][3]
1. 染料配制
储存液：避光，分装保存，避免反复冻融。
工作液：无血清培养基稀释母液到工作浓度。预热染色工作液到 37 °C。可根据实际情况稀释相应的储备液。注意，如果溶剂为 DMSO，则必须考虑 DMSO 的细胞毒性，并应制备溶剂对照；如果溶剂为纯水，则工作液在加入细胞前需要过滤并除菌。
2. 细胞标记
移除细胞培养基，用 PBS 洗涤细胞。
加入染料工作液，37 °C孵育 30 分钟。
移除染料，用预温培养基洗涤 3 次。
换新鲜培养基，继续培养或实验 (荧光信号在 1-2 小时内达峰值)。
3. 检测
荧光显微镜： (Ex 360 nm, Em 460 nm)。
