1. 溶液配置[1]
1.1 工作液的配制
用缓冲液稀释 (例如含 0.04% Poloxamer 407 (F127) (HY-D1005) 的 HHBS) 稀释 (根据实验优化)。可根据实际情况稀释相应的储备液。注意，如果溶剂为 DMSO，则必须考虑 DMSO 的细胞毒性，并应制备溶剂对照；如果溶剂为纯水，则工作液在加入细胞前需要过滤并除菌。
注意：工作液现配现用，避光。

2. 细胞染色
2.1 计数细胞，从每个样本 (对照和实验) 中取 106个细胞。
2.2 收集细胞 (5 分钟，3000×g，4°C) ，用 PBS 洗涤一次。
2.3 用 0.5 mL PBS 重悬细胞，加入 0.5 μL Fluo-4- AM 至最终浓度为 1 μM。37°C 孵育 1 小时。
2.4 用 PBS 洗涤细胞 3 次 (2 分钟，3000×g) ，最后用 1 mL PBS重悬。
2.5 用流式细胞术评估染色 (Ex/Em = 485/526 nm)。
