HADA hydrochloride (HCC-Amino-D-alanine hydrochloride) is a blue (λem~450 nm) fluorescent D-amino acid (FDAA). FDAAs are efficiently incorporated into the peptidoglycans (PGs) of diverse bacterial species at the sites of PG biosynthesis, allowing specific and covalent probing of bacterial growth with minimal perturbation[1].

1. 溶液制备[2]
1.1 原液保存
储存：分装后，避光保存于 -20°C 或 -80°C。避免反复冻融。
1.2 工作液配制
用细胞培养基稀释。可根据实际情况稀释相应的储备液。注意，如果溶剂为 DMSO，则必须考虑 DMSO 的细胞毒性，并应制备溶剂对照；如果溶剂为纯水，则工作液在加入细胞前需要过滤并除菌。
注意：工作液应立即配制并使用，并避光保存。

2. 金黄色葡萄球菌染色
2.1 将金黄色葡萄球菌接种于培养皿中。
2.2 将 250 μM HADA hydrochloride 加入细胞中。
2.3 37°C 孵育细胞 30 分钟 (根据实验优化)。
2.4 吸去染料工作液，用培养基洗涤 2-3 次，每次 5 分钟，并在荧光显微镜下观察。
