Thioflavin T (solution) is a cationic Benzothiazole dye that shows enhanced fluorescence upon binding to amyloid in tissue sections. Excitation max.: ~385 nm (free) ; ~450 nm (bound) ; Emission max.: ~445 nm (free) ; ~485 nm (bound) [1][2][3].

Thioflavin T 是一种荧光染料,可以和富含 β 折叠的结构结合,常用来测定蛋白的原纤化。一旦结合,Thioflavin T 的发射光谱就会发生红移,并且荧光强度也会增强,也可以用来检测原纤维形成。以下是 Aβ 纤维检测的参考方案[2][3]

1. 配制包含 LMW Aβ42 (5.5 μM) 、Thioflavin T 溶液 (20 μM) 和 NaCl (100 mM) 的混合样品。
2. 在 96 孔微孔板中配制混合物。密封微孔板,并将其置于 37°C 的振荡培养箱中,孵育指定时间。
3. 检测前摇晃 20 秒。将激发光波长设为 450 nm,发射光波长设为 485 nm。使用酶标仪读取荧光值。
