Cpn10 Antibody (YA2190) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to Cpn10.

Cpn10 是一种共伴侣蛋白，参与线粒体蛋白输入和大分子组装。它与 Hsp60 协同作用，促进输入蛋白的正确折叠，并可能防止错误折叠，同时在应激条件下促进线粒体基质中未折叠多肽的重新折叠和正确组装。这些伴侣蛋白的功能单元由 Hsp60 大亚基的七聚体环组成，形成背对背的双环结构。在一个循环反应中，Hsp60 环复合物每个环结合一个未折叠的底物蛋白，随后结合 ATP 并与共伴侣蛋白 Hsp10 的两个七聚体环结合。这导致底物蛋白被隔离在 Hsp60 的内腔中，使其在一定时间内不受其他细胞组分的干扰而完成折叠。所有 Hsp60 亚基中 ATP 的同步水解导致伴侣蛋白环的解离，并释放 ADP 和折叠完成的底物蛋白。

  Western blot analysis of extracts from Hela(lane 2(20ug) ,A549(lane 3(20ug) and A431 (lane 4(20ug) using Cpn10 Antibody (HY-P82445) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80438, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.

  Immunocytochemistry analysis of HELA cells labeling Cpn10 with Cpn10 Antibody (HY-P82445) at 1/50 dilution. Cells were fixed in 4% paraformaldehyde for 15 minutes at room temperature, permeabilized with 0.1% Triton X-100 for 10 minutes at room temperature, then blocked with QuickBlock™ Blocking Buffer for Immunol Staining for 10 min at room temperature. Cells were then incubated with Cpn10 Antibody (HY-P82445) at 1/50dilution in QuickBlock™ Blocking Buffer for Immunol Staining at 4 ℃. Alexa Fluor® 488-conjugated AffiniPure Goat Anti-Rabbit IgG H&L（HY-P8002, Green) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. The Nuclear counterstain was DAPI (Blue).

Cells were fixed in 4% paraformaldehyde for 15 minutes at room temperature, permeabilized with 0.1% Triton X-100 for 10 minutes at room temperature, then blocked with QuickBlock™ Blocking Buffer for Immunol Staining for 10 min at room temperature. Cells were then incubated with Cpn10 Antibody (HY-P82445) at 1/100 dilution in QuickBlock™ Blocking Buffer for Immunol Staining at 4 ℃. Alexa Fluor® 488-conjugated AffiniPure Goat Anti-Rabbit IgG H&L（HY-P8002, Green) was used as the secondary antibody at 1/1,000 dilution.   Immunohistochemical analysis of paraffin-embedded Rat testis tissue using Cpn10 Antibody (YA2190). The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 8.0) for 8 minutes. The tissues were blocked in QuickBlock for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (HY-P82445, 1/100) in 4℃ overnight. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.
