Phospho-Rb (Ser807) Antibody (YA159) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to Phospho-Rb (Ser807).

Rb (Retinoblastoma protein) 是一种关键的肿瘤抑制因子，主要调控细胞周期的 G1/S 期转换。其低磷酸化形式通过与 E2F 家族转录调节因子结合，抑制 E2F 响应基因的转录。Rb 不仅通过物理阻断 E2F 的转录激活域发挥作用，还通过招募染色质修饰酶（如组蛋白甲基转移酶 SUV39H1、KMT5B 和 KMT5C）主动抑制转录，维持异染色质的结构稳定性，尤其是组成型异染色质，并控制组蛋白 H4 'Lys-20' 的三甲基化。CDK/cyclin 依赖性磷酸化会促使 Rb 与 E2F 解离，从而激活 E2F 响应基因的转录并触发 S 期进入。此外，Rb 还能通过 CDK3/cyclin-C 的磷酸化激活促进 G0-G1 期转换。在神经元中，Rb 与 SMARCA4/BRG1 协作，通过招募 HDAC 复合体抑制 c-FOS 启动子转录；钙离子内流时，钙调磷酸酶使 Rb 去磷酸化，导致抑制复合体解离（By similarity）。在病毒感染时，SV40 大 T 抗原、HPV E7 蛋白或腺病毒 E1A 蛋白会破坏 Rb-E2F1 复合体，干扰 Rb 的功能。

  Immunocytochemistry analysis of L929 cells labeling Phospho-Rb (Ser807) with Phospho-Rb (Ser807) antibody (HY-P80476) at 1/50 dilution. Cells were fixed in 4% paraformaldehyde for 15 minutes at room temperature, permeabilized with 0.1% Triton X-100 for 10 minutes at room temperature, then blocked with QuickBlock™ Blocking Buffer for Immunol Staining for 10 min at room temperature. Cells were then incubated with Phospho-Rb (Ser807) antibody (HY-P80476) at 1/50 dilution in QuickBlock™ Blocking Buffer for Immunol Staining at 4 ℃. Alexa Fluor® 488-conjugated AffiniPure Goat Anti-Rabbit IgG H&L（HY-P8002, Green) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. The Nuclear counterstain was DAPI (Blue).

  Western blot analysis of extracts from K562 (lane 2(20μg) and K562 (lane 3(40μg)using Phospho-Rb (HY-P80476) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% BSA in TBST for 2 hour at room temperature. The primary antibody (HY-P80476, 1/1000) and Loading control antibody (Beta Actin, HY-P80438, 1/3000) was used in 5% BSA in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (HY-P8004/HY-P8001, 1/10,000) was used for 1 hour at room temperature.
