Filipin complex (solution) 

Filipin complex (solution)is a potent polyene macrolide antifungal antibiotic. Filipin complex inserts into membranes and sequester cholesterol into complexes and inhibits PRRSV entry. The Filipin complex consists of about 75.8% Filipin III (HY-N6718), 10.8% Filipin IV, 9.1% Filipin II, and 1.2% Filipin I (Ex/Em = 380/430 nm)^[1][2][3].
Solution Concentration: 5 mg/mL

1.1 工作液配制
用 PBS 或无血清细胞培养基稀释至 50 μg/mL (根据实验优化)。
注意：工作液应立即配制并使用，并避光保存。
母液分装后，避光保存于 -20°C 或 -80°C，避免反复冻融。

用于神经元胆固醇水平染色。
1. 染色切片用 PBS 洗涤三次，并与 1.5 mg/mL 甘氨酸在室温下孵育 10 分钟。
2. 然后用 0.05 mg/mL Filipin complex (溶于含 10% FBS 的 PBS 中) 染色切片 2 小时，并用 PBS 漂洗三次。
3. 使用共聚焦显微镜 (激发/发射= 380/430 nm) 进行成像。

Filipin complex (2 μg/mL；1+48 小时) 抑制 PRRSV 进入，在 MARC-145 细胞中，受感染细胞中的呼吸综合征病毒 (PRRSV) 基因组拷贝减少了 61%^[2]。
