Z-Gly-Gly-Arg-AMC is a thrombin-specific fluorogenic substrate for testing of thrombin generation in PRP and platelet-poor plasma (PPP) (Ex/Em = 390/480 nm)[1][2].

(In Vitro)

1. 溶液配制[3]
1.1 储备液配制
溶剂：水
浓度：10 mM。
储存：分装后，置于 -20°C 或 -80°C 避光保存。避免反复冻融。
1.2 工作液配制
用 PBS (或其他缓冲液) 稀释至 10 µM (根据实验优化)。
注意：工作液应立即配制并使用，并避光保存。

2.凝血酶抑制筛选
2.1 将样品和凝血酶依次转移至 384 孔板中。
2.2 孵育 15 分钟后，加入 Z-Gly-Gly-Arg-AMC (50 μM)。
2.3 使用含有 10 mM Tris-Cl (pH 7.4)、150 mM NaCl、10 mM MgCl2、1 mM CaCl2、0.1% w/v BSA、0.01% v/v Triton-X100 和 1% v/v DMSO 的缓冲液以及荧光底物 Z-Gly-Gly-Arg-AMC (50 μM) 进行测量
2.4 使用荧光读数仪 (Ex/Em = 360/465 nm) 在 25°C 下测量荧光强度，持续 30 分钟，每 3 分钟读取一次。
