Western blot analysis of extracts from A431(lane 2(20ug) and A431(lane 3(40ug) using ABCB5 Antibody (HY-P80527) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P83730, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.
Immunocytochemistry analysis of A549 cells labeling ABCB5 with ABCB5 Antibody (HY-P80527) at 1/100 dilution. Cells were fixed in 4% paraformaldehyde for 15 minutes at room temperature, then blocked with QuickBlock™ Blocking Buffer for Immunol Staining for 10 min at room temperature. Cells were then incubated with ABCB5 Antibody (HY-P80527) at 1/100dilution in QuickBlock™ Blocking Buffer for Immunol Staining at 4 ℃. Alexa Fluor® 488-conjugated AffiniPure Goat Anti-Mouse IgG H&L（HY-P8005, Green) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. The Nuclear counterstain was DAPI (Blue).

背景

功能：能量依赖性外排转运蛋白负责降低多药耐药细胞中的药物蓄积 (PubMed:12960149, PubMed:15205344, PubMed:15899824, PubMed:22306008)。该蛋白特异性存在于角膜缘干细胞中，并在角膜发育和修复中发挥关键作用 (基于相似性)。

亚细胞定位：细胞膜；多通道膜蛋白

表达水平：
组织特异性：在表皮黑色素细胞中，CD133 表达的祖细胞表达该蛋白 (蛋白水平) 。该蛋白广泛表达，并在色素细胞中特异性表达。在多种恶性组织中高表达：在临床黑色素瘤中高表达，在正常皮肤中低表达。在黑色素瘤中，该蛋白标记恶性黑色素瘤起始细胞 (MMIC) ，这些细胞具有无限的自我更新能力，导致肿瘤不可逆转地进展和转移，从而引发临床恶性肿瘤。该蛋白也在多种白血病细胞中高表达。在基底角膜缘上皮中也有表达。
