DOTAP methylsulfat is a cationic lipid reagent, a cationic derivative of trimethylammonium, linked to two 18-carbon fatty acid tails, each with a single olefin group. DOTAP methylsulfat can self-assemble with negatively charged ions (such as DNA) to form complexes, which can be adsorbed to the cell membrane surface and enter the cell by electrostatic interaction and endocytosis, respectively. DOTAP methylsulfat promotes endosomal membrane fusion with its own hydrophobic domain, releases DNA into the cytoplasm, and exerts gene delivery function. DOTAP methylsulfat can be widely used in research fields such as gene therapy, cell transfection, and non-viral vector design[1][2].

(In Vitro)

DOTAP methylsulfat (不同质量比；4 小时转染+24 小时培养) 与 DOPE (HY-112005) 的不同质量比具有不同的最佳转染效率。在 Huh7 和 AGS 细胞中，最佳配比为 T1P0 (1:0)/T3P1 (3:1) 型，在 A549 细胞中，最佳配比为 T1P1 (1:1)/T3P1 (1:3) 型，且与脂质体尺寸或 zeta 电位无关[1]。
DOTAP methylsulfat 于脂质按一定比例 (如：1:2，1:1，2:1) 混合，比单独使用 DOTAP methylsulfat 转染 DNA 的效率更高，在 80% 血清浓度下仍保持转染活性。并且 DOTAP methylsulfat 与 DNA 的最佳转染比例为 12:1[2]。
DOTAP methylsulfat (30 分钟 DNase 处理) 在血清存在时，DNA- 脂质复合物对 DNase I 消化保持稳定，胆固醇的加入不影响复合物结构完整性[2]。
DOTAP methylsulfat (含 1% NBD-DOTAP methylsulfat 标记；2 小时转染) 在流式细胞术分析中，胆固醇修饰的脂质体在血清中与细胞结合的数量显著高于纯 DOTAP methylsulfat 脂质体[2]。

——为保证实验结果的可靠性，澄清的储备液可以根据储存条件，适当保存；体内实验的工作液，建议您现用现配，当天使用；
以下溶剂前显示的百分比是指该溶剂在您配制终溶液中的体积占比；如在配制过程中出现沉淀、析出现象，可以通过加热和/或超声的方式助溶

  • 方案 一

    请依序添加每种溶剂： 10% EtOH    90% Corn Oil

    Solubility: ≥ 2.5 mg/mL (3.23 mM); 澄清溶液

    此方案可获得 ≥ 2.5 mg/mL（饱和度未知）的澄清溶液，此方案实验周期在半个月以上的动物实验酌情使用。

    以 1 mL 工作液为例，取 100 μL 25.0 mg/mL 的澄清 EtOH 储备液加到 900 μL 玉米油中，混合均匀。

  • 方案 二

    请依序添加每种溶剂： 10% DMSO    40% PEG300    5% Tween-80    45% Saline

    Solubility: ≥ 1.67 mg/mL (2.16 mM); 澄清溶液

    此方案可获得 ≥ 1.67 mg/mL（饱和度未知）的澄清溶液。

    以 1 mL 工作液为例，取 100 μL 16.7 mg/mL 的澄清 DMSO 储备液加到 400 μL PEG300 中，混合均匀；再向上述体系中加入 50 μL Tween-80，混合均匀；然后再继续加入 450 μL 生理盐水 定容至 1 mL。

    生理盐水的配制：将 0.9 g 氯化钠，溶解于 ddH₂O 并定容至 100 mL，可以得到澄清透明的生理盐水溶液。
