Muromonab  (Synonyms: 莫罗单抗; Muromanab-CD3)

Muromonab (Muromonab-CD3; OKT3) is a mouse monoclonal antibody targeting the CD3 antigen. Muromonab specifically binds to the CD3 antigen on the surface of human and higher primate T cells. Muromonab blocks the function of T cell receptors to recognize foreign antigens and inhibits T cell-mediated immune responses, including cell-mediated lymphocyte lysis and T cell proliferation responses. Muromonab can be used to study acute kidney, liver, heart and combined kidney-pancreas transplant rejection, and can also be used to study graft-versus-host disease in bone marrow transplant patients^{[1][2][3][4][5]}.

Muromonab 可阻断细胞毒性 T 细胞功能并抑制 T 细胞增殖^[1]。
Muromonab 与 T 细胞上的 CD3 抗原结合，导致体外循环中 CD3⁺ T 细胞被快速清除/耗竭，减少 CD4⁺ 和 CD8⁺ 亚群^[1]。

Muromonab 还诱导 IL-2 和 IFN-γ 等细胞因子的释放，从而促进早期免疫激活标志物的产生^[1]。Curcumin (HY-N0005) 和 Sirolimus (HY-10219) 分别呈浓度依赖性抑制 Muromonab 诱导的人外周血单核细胞 (PBMC) 增殖 (IC₅₀=2.8 μM 和 1.1 ng/mL)，两者低浓度联用具有协同作用^[6]。

Muromonab 在猕猴肾移植模型中显著延长移植肾存活时间并抑制急性排斥反应^[1]。
Muromonab (5 mg；静脉注射；2-3 次) 的平均稳态血浆谷浓度约为 0.9 mg/L，此时的血浆浓度具有阻断细胞毒性 T 细胞的功能^[1]。

  Flow cytometric analysis of 10⁶ Jurkat cells labeling Muromonab (HY-P99152, red). Cells were fixed with 4% paraformaldehyde. Then stained with the primary antibody at 1/200 dilution for an hour at 4℃. Alexa Fluor® 488-conjugated AffiniPure Goat Anti-Mouse IgG H&L (HY-P8005) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Mouse IgG2a (HY-P99978, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).
