WSP-1 is a selective and rapid-reaction H2S specific fluorescent dye (Ex/Em=465/515 nm). WSP-1 reacts with H2S with the releasing of fluorophore[1].

Guidelines (Following is our recommended protocol. This protocol only provides a guideline, and should be modified according to your specific needs).
细胞内 H2S 的荧光检测 (荧光显微镜检查)[1]:
1. HASMCs 培养至约 90% 聚集，实验前 24 小时，将细胞以 30*103 每孔的密度在 8 孔培养载玻片上培养 24 小时。
2. 更换培养基，加入 WSP-1 工作液 400 μL，37℃ 孵育 30min。
3. 将 WSP-1 工作液替换为 380 μL 标准缓冲液，加入 20 μL 所需浓度的H2S 供体化合物。
4. 当 WSP-1 与 H2S 反应时，释放在 Ex/Em=465/515 nm 下可检测到的荧光团。取缓冲标准品，培养 1 小时后用 400 μL DPBS 洗涤。
5. 去除 DPBS，室温下每孔加入 200 μL Bouin 溶液固定细胞 10 min。去除多余的Bouin溶液，用 400 μL DPBS 清洗细胞 2 次。
6. 用荧光显微镜检查和获取荧光信号。

  • Do most proteins show cross-species activity?
