COX-2-IN-59 is a potent and selective COX-2 inhibitor with an IC50 of 0.052 μM. COX-2-IN-59 exhibits 200-fold selectivity over COX-1 (IC50 = 11.16 μM). COX-2-IN-59 reduces COX-2 levels, induces cell cycle arrest, and triggers apoptosis by increasing Bax expression, decreasing Bcl-2 levels, and activating caspase-3. COX-2-IN-59 can be used for the research of colon cancer[1].

Cell Viability Assay[1]

Cell Line: A-549, HOS, MCF-7, HT-29, PC3, and Caco-2 cells
Concentration: 12.5-100 μM
Incubation Time: 24 h
Result: Inhibited A-549, HOS, MCF-7, HT-29, PC3, and Caco-2 cell growth with IC50s of 4, 67.3, 30.5, 8.5, 29.1, and 2.6 μM, respectively.

Apoptosis Analysis[1]

Cell Line: Caco-2
Concentration: 2.6 μM
Incubation Time: 24 h
Result: Demonstrated potent proapoptotic activity in Caco-2 cells, inducing a nearly eightfold increase in total apoptosis (24.38% vs. 3.16% in controls). Induced a dramatic 63-fold increase (15.86% vs. 0.25%) in late apoptosis, and an 11-fold increase (4.85% vs. 0.42%) in early apoptosis.

Cell Cycle Analysis[1]

Result: Showed a dramatic decrease in G2/M phase cells in the treated group (3.82%) compared to the control group (22.05%). Significantly increased cells in the G0/G1 phase (64.06% vs. 48.29% in control). Induced G1/S phase cell cycle arrest.
